To cope with their environment, plants synthesize a wide range of natural products, such as the monoterpene indole alkaloids (MIAs) produced by species of the order Gentianales, including the well-known Madagascar periwinkle and devil pepper. Many of these MIAs display highly valuable biological activities, as exemplified by the anticancer drug vinblastine and the antimalarial compound quinine. Over the years, the quest to elucidate MIA biosynthetic pathways has mobilized numerous research groups worldwide. In this presentation, I will review the recent history of MIA gene discovery, highlighting the step-by-step development of an efficient gene identification pipeline—from raw transcriptomic analyses and high-quality genome sequencing to single-cell RNA sequencing. Finally, I will show how the transfer of these genes into yeast to create cell factories may pave the way for a sustainable supply of MIAs.
